Orexin Receptor Antagonists for Insomnia
Where behavioral interventions are not effective, pharmacological treatments can offer an alternative for insomnia patients. However, while benzodiazepines and "Z-drugs" can be effective in the short term, long-term use is associated with side effects including the development of tolerance and addiction. Orexin receptor antagonists, a relatively new class of medications, have reduced dependance and tolerance-inducing effects, showing promise for longer-term use. Given the emerging importance of orexin receptor antagonists in managing insomnia, Nature and Science of Sleep here collects original research articles, reviews, and perspectives exploring the clinical efficacy, safety, mechanisms of action, and real-world application of these therapies.
